Anthocyanins in the sepal of Hibiscus sabdriffa L. is a kind of flavonoid and show the antioxidant effect. In this study, four dosages (240, 260, 280, and 300 Gray of gamma irradiation) were applied to the seeds of Hibiscus sabdriffa L. for inducing random mutation. Anthocyanins were extracted from the mature sepal of first generation seeds, separated by chromatographic filter paper, and the content of anthocyanin of the sepal of Hibiscus sabdriffa L. mutants and the quantified the delphinidin and cyanidin were estimated by HPLC. The result showed that anthocyanin contents of M1 mutants, C280-108, 260-12, 260-15, 280-6, C 280-19, and C-280-34 increased significantly. The M3 generation anthocyanins content of C280-180 and 260-12 were higher than control for 51% and 28% respectively. However, germination vigor of seeds of mutant tend to be decrease. The free-radical scavenging capacity of extract of Hibiscus sabdriffa L. was measured and showed positively related with anthocyanin content. The antioxidant systems of free-radical scavenging in the organism include enzymatic defense system, such as superoxide dismutase, and non-enzymatic defense system, such as the antioxidant as ascorbic acid and tocopherol. In this experiment, we analyzed the distribution and the activities in the Hibiscus sabdriffa L. leaves and sepals. The SDS-PAGE analysis and SOD activity electrophoresis were compared and found that the extract of Hibiscus sabdriffa L. sepal did not have the superoxide dismutase activity and the ability of scavenging O2․—of sepal was due mainly to anthocyanins. The abilities against lipid peroxidation were measured to analyze the antioxidant activities of Hibiscus sabdriffa L. sepal. Anthocyanins scavenge lipid peroxide free-radical to decrease lipid peroxide free-radical to attack the polyunsaturated fatty acid of bio-membrane to lower the production amount of lipid peroxidative reactant MDA. The result showed that the extract of sepal had capacity to inhibit lipid peroxidatiion. Two mutants, C280-108 and 260-12 with enhanced anthocyanins content were obained by γ- ray irradiation, suggests that this method may contribute to increasing the antioxidant effect of Hibiscus sabdriffa L.
