Preadipocytes (3T3-L1) and myoblasts (C2C12) cell line of rats are used in this experiment, which are divided into the control group, Pic group (200ppb picolinate) and CrPic group (200ppb chromium picolinate) at random. The result of differentiation of preadipocyte shows that in the differentiating adipocyte and their triglyceride content in CrPic group is manifestly lower than the control group (p <0.05). Compared with control group, expression of transcription factor of peroxisome proliferator-activated receptors γ (PPARγ) in CrPic and Pic groups are manifestly depressed (p <0.0001). The activity of glycerophosphate dehydrogenase (GPDH) in CrPic and Pic groups are depressed, and the result of CrPic group is largely different with that of the control group (p <0.0001). When compared with the control group, the activity of fatty acid synthetase (FAS) in CrPic group is evidently repressed as well (p <0.05). But there is no significantly different in the activity of malic dehydrogenase (MDH) among the three groups (p >0.05). In the aspect of proliferation of preadipocytes, the cells in CrPic is lower than that in the control group (p <0.05), but the DNA content in three groups is no significantly different (p >0.05). The result of differentiation of myoblasts reveals that there is no markerly different in the number of differentiated myotubes, activity of creatine kinase (CK) and aldolase (ALB) among the three groups (p >0.05). The activity of hexokinase in CrPic and Pic group is obviously higher than that in the control group (p <0.05), and the activity of myokinase in Pic group is higher than that in CrPic and the control group (p <0.05). Besides, there is no effect on the transcription factor myogenin among the three groups (p >0.05). In the aspect of proliferation of myoblasts, there is no influence on the numbers of proliferation of the three groups (p >0.05), and the amount of DNA in the control group is higher than that in Pic group (p <0.05). This experiment indicated that 200ppb chromium picolinate can significantly depress proliferation and differentiation of preadipocytes; while no effective on the proliferation and differentiation of myoblasts.
