Recently, its population has gradually decreased due to man-made destruction to the plant and its habitats. How to recover its population has become an important issue. In this research, the techniques for propagation and cultivation of L. longiflorum var. formosanum were developed first and then its seeds and seedlings were inoculated with arbuscular mycorrhizal fungi (AMF) to study the effect of AMF on the growth of the plant. Field investigations showed that some of the L. longiflorum var. formosanum population was still spotted in the wild in Taiwan. Its flowering season was affected by three critical factors ─ elevation, temperature and shading. It reguired only 4 months to flower when seed sowed in June. The suitable temperature range for the germination of the seeds collected in the restoration farm in Fu-Hsing Village was 10℃~25℃ and the germination rates were as high as 100%. The seeds stored at 10 ℃ had a low germination rate at the beginning, but it could reach 95% in the final stage of the storage. The germination rate of the seeds collected from their original habitats, Tien-Chih, reached 100% under 15~20℃ for 20 days. This rate could be maintained when seeds were stored in the range of 4 ~10℃. However the rate decreased when seeds were stored under the room temperature for 6 months. This decrease continued until it reached 0% after 10 months. Seeds from the brown or yellow-brown capsule had 100% germination rate under 15~20℃ after 20 days from sowing. Seeds and seedlings of L. longiflorum var. formosanum were inoculated with different species of AMF. The results showed that Glomus clarum, G. etunicatum, G. fasiculatum, G. mosseae and Acaulospora sp. significantly enhanced the development of bulb circumference. The results also showed that G. clarum, G. fasiculattum and G. mosseae significantly increased the fresh weight of the plant.
