Description
Summary:碩士 === 中國文化大學 === 生活應用科學研究所 === 93 === Abstract The aim of this study was to evaluated the antioxidant substances, antioxidant capacity and the protection of DNA from oxidative damage in lymphocytes induced by H2O2 of acidic methanol extract and aqueous extract of nine herb plants. The herb plants included Morus alba L., mulbrry tea, Bidens pilosa L. var. minor., Lycium chinense mill., sikworm faeces tea, Mentha piperita., Plantago asiatica Linn., Houttuynia cordata Thunb and Centella asiatica (L) Urban.. The antioxidant substances such as flavonol and polyphenol were determinated. The antioxidant capacity was evaluated by the inhibition percentage of conjugated diene formation in linoleic acid emulsion system and by the TEAC ( trolox equivalent antioxidant capacity ) value. The DNA oxidative damage was evaluated by comet assay. The correlation between antioxidant substance, antioxidant capacity and the protection of DNA from oxidative damage in lymphocytes were analyzed. In results, the herb plants were rich in flavonols. Especially, B.pilosa L. var., minor., C. (L) Urban. and M. piperita., rich all in myricetin. The M. piperita., C. asiatica (L) Urban. and B. pilosa L. var. minor. abundant in morin, The H. cordata Thunb, silkworm faeces tea and M. alba L. abundant in quercetin. The C. asiatica (L) Urban., silkworm faeces tea and M. alba L. abundant in kaemperol. The polyphenol content was rich in sikworm faeces tea, B. pilosa L. var. minor., C. asiatica (L) Urban., L. chinense mill. and P. asiatica Linn. In the conjugated diene system, the acidic methanolic extracts of P. asiatica Linn., H. cordata Thunb, C. asiatica (L) Urban. and M. piperita. had the best inhibition percentage. In TEAC system, the aqueous extract of H. cordata Thunb, B. pilosa L. var. minor. and silkworm faeces tea had the best inhibition percentage. There was significantly positive correlation between quercetin and the TEAC value. In comet assay, the acidic methanolic extracts of L. chinense mill., M. piperita., P. asiatica Linn., H. cordata Thunb. and C. asiatica (L) Urban., significantly inhibited lymphocytes from DNA oxidative damage. The aqueous extracts of herb plants, M. piperita., P. asiatica Linn., H. cordata Thunb and C. asiatica (L) Urban. significantly inhibited lymphocytes from DNA oxidative damage. In conclusion, the herb plants rich in flavonols had strong antioxidant capacity and were able to protect DNA of lymphocytes from oxidative damage. Particularly, H. cordata Thunb rich in quercetin and P. asiatica Linn. rich in polyphenol had the best effect. Key words: Herb plants, Antioxidant, Flavonols, Conjugated diene, TEAC, Comet assay.
