Summary:碩士 === 實踐大學 === 食品營養研究所 === 92 === For understanding the contents of needle of daylily’s flavonoids and colchicine, the needle of daylily’s was extracted by different solution (methanol, ethanol, chloroform, dichloromethane), the extracted thing was analized through spectrophotometer to measure the contents of total phenols、flavonoids、carotenoids、chlorophylls 、DPPH (1,1-dipheny-2-picrylhydrazyl) radical activeity、ferrous ion chelating power and the ability of reducing potassium ferricyanide. The result shows that the quantity of total phenols、carotenoids、chlorophylls of one day daylily’s were obviously higher than three days daylily, but in the part of flavonoids, were contrary different. The capture ability at 1000 ppm of methanol extracted solution was about 60-80%, scavenging the chelating ferrous ion power also strong at 2000 ppm was about 60-80%, but the reducing power is lower. The category and quantity of the needle of daylily’s flavonoids were different area and flower time. The needle of daylily was extracted by 95% ethanol the quantity of Rutin, Daidzein and Quercetin in three days daylily are higher, there are 186 ppm, 161 ppm and 157 ppm respectively. The quantity of the Quercetin of the three days flower which from Hualien is the higher, about the 189 ppm. The quantity of the Myricetin and Quercetin in two days flower which from Hualien is the higher, about the 471 ppm and 325 ppm. The quantity of the Kaempferol, Myricetin and Quercetin in one day flower which from Hualien is higher, about 2207 ppm, 323 ppm and 152 ppm individualy. The needle of daylily was extracted by different solution suchas methanol, ethanol, chloroform, dichloromethane were analized through TLC, HPLC and LC/ MS/ MS. Although it showed at the TLC with the yellow spot, but can not found significant peak in the spectrum of HPLC. Eventually, the different column were used, but there was not any obvious and same retention time was retened as colchicine. Finally, analyzing with LC/ MS/ MS, the result showed fragments were not alike to standard in which the methyl alcohol extracted rolution was not contain the colchicines in needle of daylily.
