Twenty-four female SD rats were weight-matched in to three groups, including control group (C), 34℃ water temperature training group (W), and 25 ℃ water temperature training group (CO). After two weeks of swimming training, the muscle GLUT4 protein gene expression, glycogen storage, and whole body glucose tolerance were studied. Our results showed that glycogen content of red and white quadriceps muscles in CO and W groups were significantly higher than control group. Simultaneously, the glycogen content of red and white gastrocenimus muscles also revealed the consistent pattern as quadriceps muscles. Moreover, the GLUT4 protein and gene expressions of red and white gastrocenimus muscles were significantly higher in both warm and cold temperature exercise training groups. But, interestingly, we did not find any statistical differences in muscle GLUT4 gene expression between both training groups. In conclusion, we demonstrated that swimming training in 25℃ water can significantly enhance glycogen storage but not through the alteration in muscle GLUT4 protein expression.
