Dermatogoides pteronyssinus(Der p)is one of the most prominent and important species of house dust mite in allergic asthma in Taiwan. We used Der p to induce BABL/c mice as asthmatic animal model which help us to understand the therapeutic mechanism of you- gui- wan(YGW). In this study, we found that YGW could inhibit the recruitment of monocyte and eosinophil in brochoalveolar lavage fluid (BALF). Simultaneously, it could reduce CD3+CD4+and CD4+CD25+ T cell in lymphacyte ratio significantly. YGW also could down-regulated IL-5 in serum to inhibit eosinophil infiltration and up-regulated IL-12 in BALF to change Th2-bios. Otherwise, EMSA and immunohistochemistry stain demonstrated YGW could inhibit NF-κB expression in lung. Collagen assay and hematoxylin-eosin stain indicated YGW could reduce airway remodling in airway. In RT-PCR experiment, we also found that YGW could inhibit TGF-β1, IL-10, IL-13, eotaxin, RANTES, and MCP-1 mRNA expression which demonstrated that the major mechanism of YGW still inhibited late asthmatic response.
