Previous studies have demonstrated that oolong tea has anti-fungal and antioxidant activities. However, the whitening and antioxidant capacities of oolong tea temperature-dependent extracts have not been explored. In this study, the effects of Oriental beauty tea water extracts on mushroom tyrosinase activity, B16F10 murine melanoma cellular tyrosinase activity and cellular melanin content were determined spectrophotometrically. The expression of tyrosinase and related proteins in B16F10 cells were determined by Western blotting. Our data indicated that Oriental beauty tea water extracts reduce mushroom tyrosinase activity ,also decrease the amount of melanin and effectively suppressed intracellular tyrosinase activity in B16F10 murine melanoma cells. In addition, Oriental beauty tea water extracts also effectively decrease intracellular reactive species (RS) and reactive oxygen species (ROS) levels at the same dose range. Our results revealed that Oriental beauty tea water extracts inhibit melanogenesis in B16F10 cells by down- regulation of tyrosinse and Microhptalmia-associated transcriptional factor(MITF) expression or through its antioxidant properties. Hence, Oriental beauty tea water extracts could be used as a good skin whitening or antioxidation agent. Key words: Oriental beauty tea , B16F10, tyrosinase, melanin, MITF
