A transition-state analogue, N-?t-BOC-L-phenylanine-2-aminopyridine, of the target substrate will be acylated to generate a surfactant-like structure, N-?decyl-l-phenylalanine-2-aminopyridine, and then be employed as a template molecule. To study the possibility of preparing high-crosslinked polymer particles by inverse suspension polymerization, span-60 was employed as the surfactant for model study. Three commercially available crosslinking agents, N, N’-methylenebisacrylamide, N, N’-ethylenebisacrylamide and 1,4-diacryloylpiperazine were studied at room temperature. For N, N’-methylenebisacrylamide, particles with 10 mole% crosslinking agents can be prepared at the stirring rate of 600 and 800 rpm. For N, N’-ethylenebisacrylamide and 1,4-diacryloylpiperazine, particles with 10 mole% crosslinking agents can be prepared at a higher stirring rate of 1000 rpm. For N, N’-ethylenebisacrylamide, the percentage of crosslinking agents can be further increased to 20% by the addition of methanol to the water phase (25% v/v). Since we are interested in the preparation of high-crosslinked particles, the preparation temperature are increased to 55℃. N, N’-ethylenebisacrylamide are used as crosslinking agents because it has the highest aqueous solubility. Particles with 50 mole% crosslinking agents are prepared successfully. Particle formation is affected by the stirring rate and the concentration of span-60. Particles with the sizes of 17±5μm and 22±5 μm for 30 mole% and 50 mole% crosslinking agents respectively can be prepared at the stirring rate of 800 rpm and at the concentration of 9.7 mM/l . Further increase the stirring rate to 1000 rpm or decrease to 400 rpm result in some bulky structure.
