Summary:碩士 === 高雄醫學大學 === 醫學研究所碩士班 === 93 === Human cytomegalovirus (HCMV)is a ubiquitous herpesvirus ,which establishes a lytic infection cycle and subsequently retains a lifelong latent infection in the host. HCMV is notable for it is the leading causative pathogen for congenital cytomegalic inclusion disease a severe condition in central nerve system. After HCMV infection, a specific virion-associated protein pUL76 is transported to nucleus through inoculum. Furthermore, the evidence demonstrates that constitutively expressing pUL76 at the immediate-early phase of the viral replication cycle leads to the inhibition of HCMV production. For the purpose to elucidate the mechanisms of inhibition of viral replication and repression of gene expression at molecular and biochemical levels, yeast two-hybrid screening (Y2H)was employed to bait the cellular target proteins of UL76. In this experiment the fetal brain cDNA was used. After the library screeing,the putative cellular proteins interacting with UL76 mainly function in the ubiquitin-proteasome system(UPS) and the major cellular targeting protein of UL76 is TBP1 of 26S proteasome.
