All the three standards and the three extracts of this natural product, extracted by three different solvents (H20, ethanol and ether) with varions degree of polarity/hydrophilicity, were all tested during the optimization of separation methods (TLC and HPLC) developing. Both reverse phase and normal phase columns were used as the stationary phases for the HPLC method developing. It has been concluded that a normal phase column (VARIAN, Microsorb-MV 100-5 Si, 250x4.6mm) with 97% n-heptane: 3% ether as mobile phase is the best optimized system for following these three molecules especially the toxic compound: Emodin in the Polygonum Multiflorum Thunb. and the other molecules before and after detoxification process. Then the real tedious process simulating the Chinese herbal medicinal detoxification (Nine steaming, Nine sun-exposure and Nine cooling) was carried out. The most suitable HPLC system developed and optimized as abovementioned was used to follow the amount change of toxic substance (Emodin molecule mainly) in Polygonum Multiflorum Thunb. It was found the amount of Emodin was reduced more than half of the original amount in this natural product after the whole cycle of detoxification. Each amount of Emodin in 0, 2, 4, 6, 8 and 9 days was also analyzed and plotted and it showed an interesting point, i.e. the Emodin amount was actually increased to a peak during 2 to 4 days and decreased sharply after 4th day, then remained constant to the end of the detoxification (9 days total). This may be one of the reasons that the experienced Chinese herbal medicinal processors design the 9 days’ detoxification to make sure this folk medicine to be taken on the safe side. 吳海萍 2005 學位論文 ; thesis 94 zh-TW
