Pathological and Molecular Diagnosis on Primary Lymphoid Organ of Broiler Chickens with Emphases on Viral Immunosuppressive Diseases

碩士 === 國立屏東科技大學 === 獸醫學系 === 93 === Viral immunosuppressive diseases are commonly encountered in broiler chickens and usually lead to substantial economic losses. In addition, enhancement of secondary infections and failure of vaccinations are also frequently found in the immunocompromised chickens. The viral immunosuppressive diseases of broiler chickens manily include : Marek’s disease, chicken infectious anemia, infectious bursal disease, Newcastle Disease, pale bird syndrome and avian influenza. This study is to perform pathological and molecular diagnosis on primary lymphoid organs of the infected broiler chicken. Grossly, all the chickens have abnormal bursa and thymus. Depletion or necrosis of lymphocytes is the remarkable lesion, except that infiltration of lymphoid tumor cells is found in the cases of MDV infection. Two sets of multiplex polymerase chain reaction are developed for the simultaneous detection and differentiation of the immunosuppressive viruses. The first set is used to detect DNA viruses ( MDV and CIAV ) and the second is designed for RNA viruses ( NDV, AIV, REOV and IBDV ). In this study, DIG-labled riboprobes for the above pathogens are used to detect the sites of the agents in tissue sections. Postive signals are found in the cloacal bursa and thymus of all the cases examined, but only IBD, MD and CIA have signals in the bone marrow.
