Yin-Chen-Hao-Tang (YCHT) decoctions have been recognized as a ’’magic bullet” for jaundice and as a hepatoprotective agent for various types of liver diseases. Therefore, we used an experimental animals model of biliary atresia to test whether YCHT plays a regulatory role in the pathogenesis of hepatic apoptosis. Methods and Results:Experimental cholestasis-induced liver apoptosis after common bile-duct ligation (BDL) in rats. After surgery, YCHT (250, 500 mg/kg BW) oral administration once a day continued for 27 days. BDL caused a prominent liver collagen deposition that was supported by the increased -SMA protein and mRNA expression of TGF-1 and procollagen I. YCHT improved fibrosis in vivo. Additionally, YCHT down-regulated hepatic bax, cytochrome c protein levels and decreased in caspase 3, 8 activities of BDL rats. On the other hand, the normalizing effect of YCHT on the bax and bcl-2 mRNA expression was dependent on the YCHT dose, the dose 500 mg/kg most effectively changed the quantitative composition of both bax and bcl-2 of mRNA levels. Conclusions:These results give grounds to develop underlying studies to assume that one of the mechanisms of hepatoprotection by YCHT in BDL rats may realized via decreased apoptosis related factors activation and further mechanisms need to be elucidated.
