The effects of 6-gingerol on the inducible nitric oxide synthase (iNOS), and tumor necrosis factor-alpha (TNF-alpha) were determined in Raw264.7 cells as part of the studies on its anti-inflammatory effects. Expression of the iNOS was assessed by Western blot analyses. NO production was monitored by Griess reagent detection. 6-gingerol inhibited the induction of iNOS protein, TNF-alpha and nitrite by lipopolysaccharide (LPS) (50 ng/mL) as well as suppression of the iNOS mRNAs. 6-gingerol (20-80 microM) inhibited LPS-inducible PKC-alfa translocation to cell membrane, which was accompanied by inhibition of mitochondria membrane potential and Ca+2 mobilization. These results demonstrate that 6-gingerol inhibits LPS-inducible iNOS expression in macrophages possible through suppression of cytosolic factors which may constitute anti-inflammatory effects. In the current study, we will establish a biological approach to identify the relative anti-inflammation mechanisms of 6-gingerol. This study is valuable for the understanding the global and specific role of 6-gingerol for the development of new therapeutic modality for new drug in the future.
