Summary:碩士 === 中山醫學大學 === 口腔醫學研究所 === 94 === ZAK,the leucine-zipper(LZ) and sterile-αmotif (SAM) kinase. One of the Mixed-lineage kinase(MLK) family. It contains a kinase domain, a leucine-zipper(LZ) and a sterile-α motif (SAM) belonging to the MAP kinase kinase kinase (MAP3K)and could activate the JNK/SPAK pathway thought MKK7 in cell arrest .The expression of ZAK in cardiomyoblast cells causes dramatic change in cell morphology and induces cardiac hypertrophy. By using microarray technology ,we found Frizzled-1 gene( a receptors for the WNT family of secreted glycoproteins) will increase in ZAK over-expressing H9C2 cells. Frizzleds, cell surface receptors that mediate the actions of WNT ligands and also play essential roles in development, including cell fate, adhesion, polarity, migration, and proliferation, and many other processes in developing and adult organisms .As we know, Frizzled family includes ten member(from Frizzled-1 to Frizzled-10),Frizzled-1 couples via G-proteins Gα and Gq to the canonical β-catenin-Lef-Tcf pathway. We used NorthernBlot and RNAi technology to prove that the relationship of ZAK and Frizzled-1. We found that Frizzled-1 RNA increases when ZAK over-expression and decreases when ZAK be blocked by ZAK RNAi .We confirm the correctness of microarray data about Frizzled-1.
