We combined alginate and chitosan in preparing the 3D scaffold, and measured the growth of human gingival fibroblasts and human skin fibroblasts in the scaffold. On the other hand, we modified chitosan by incorporating 5 nm gold nanoparticles、silver nanoparticles and FGF1, tested the fibroblasts growth and extra-cellular matrix secretion such as Type I collagen content and GAGs on the films. We also analyzed the actin stress fibers, focal adhesion proteins, cell cycle, gene expression on the nanocomposites films, it was found that on chitosan-Au 40 ppm, chitosan-Ag 40 ppm and chitosan–FGF1 20 ng have the best growth and extra-cellular matrix secretion. When this formula was used to prepare alginate-chitosan 3D scaffold, the same result was found as those observed with the 2D films.
