Second, the differences of the force-distance curves between the erythrocytes extracted from different rats blood were compared. Third, the differences between the force-distance curves of the erythrocytes measured in different concentrations of saline were compared. After poly-L-lysine was used to fix erythrocytes on glasses substrate, AFM was used to measure the force-distance curve of specific erythrocytes in saline with contact mode. The force-distance curve of the erythrocytes extracted from three different rats blood in this experiment. These erythrocytes were divided into two groups, A and B. In group A, the fifteen erythrocytes were selected from each rat blood which was operated in normal saline. The erythrocytes of group B were selected from group A-3 and operated in hyposmotic saline (concentration 1.3%). The force distance curve of fifteen erythrocytes of group B were measured by AFM. From the experimental results, that measured error of AFM force distance curve measurement would be occurred due to the static electricity in the ionic solution. In order to reduce the disturbances, the suspended particles in red blood cell solution should be removed before setting up on AFM. According to the slope of the force distance curve of erythrocytes, we could obtain the mechanic property of erythrocytes. In this research, the elastic modulus can be obtained from the slop of force-distance. Then the average slope of the force-distance curve individually were A-1:-0.03233N/m、A-2:-0.02166N/m、A-3:-0.01852N/m、B:-0.01339N/m.
