To provide a helpful method for solving the hair disorders of human beings, this study tries to figure out a best solution, which is found out to be the combinations of the two growth factors, IGF-1 and SCF, and/or minoxidil. The two growth factors have been reported to be very associated with the proliferation and migration of the keratinocytes and melanocytes. And minoxidil has been used as the popularly commercial product to therapy hair baldness for over 20 years. In this study, there are two models to prove our hypothesis that the combination of the three compositions has the dual therapeutic effects on hair losing and graying. One is the in vitro culture model of hair follicles to study their effects on the hair growth. The other is the topical delivery system of liposome to detect their effects on the melanin synthesis as well as the hair growth. In the modified mini-organ culture model for hair follicles, the different combinations of IGF-1, SCF and/or minoxidil were additionally put into the control medium. The hair follicles from mice’s cheeks were carefully isolated by fine forceps and then cultured in the different treated medium. After cultured for 12 days, we took the pictures under microspore with CCD. The follicular image and hair growth-length were measured by Northern Eclipse image system of computer software. The analyzing data showed that the combination of the two growth factors, IGF-1 and SCF, had the same efficiency to stimulating hair growth as minoxidil alone. When the two growth factors are combined with minoxidil, it has the more efficiency for the hair growth than the others (P<0.05). In order to confirm the penetration ability of the two growth factors, IGF-I and SCF, to hair follicles by the topical delivery system, we developed a formula of liposome to package these proteins. It showed that growth factors within liposome could successfully penetrate into dermis through the topical application on skin. After finishing the previous proof, the two growth factors of IGF-I and SCF were first encapsulated within liposome and then mixed with the 2 % minoxidil gel as the mixture by the topical delivery system, which was tested in the graying animal model (F1 generation, C3H x Balb/c hybrid). After the local application by the mixture for 10 days, we found the faster hair growth rate in the treated mixture than in the control of PBS liposome. After the local application by the mixture for 2 weeks, mice were shaved the dorsal hair for the melanin analysis. The results indicate that the mixture is beneficial for not only the hair growth but also the melanin regeneration. In the study, it would be expected to provide a dual useful therapy for both of human hair balding and graying.
