| Summary: | 碩士 === 中國醫藥大學 === 醫學研究所碩士班 === 95 === Follicle - stimulating hormones (FSH) have been testified inducing folliculogenesis and endometrium proliferation. In this experiment, we try to find whether different injection position and frequency of recFSH will affect the number of ovary antral follicles and endometrium of the uterus. In the first experiment, immature female rats were treated twice daily(8 IU/8 times) with recFSH into abdomen, and twice daily(8 IU/8 times) or daily(4 IU/4 times) or daily(8 IU/8 times) or every other day (8 IU/2 times) or only times (8 IU/1 times) with recFSH into cloaca for four days. In the second experiment, immature female rats were treated with recFSH into abdomen or cloaca and supplemented hCG after sacrificed 18 hs. Compared with control, the total follicles of control is not increasing apparent, but injected in abdomen or cloaca will increase number of larger follicles(≧575μm) and Corpus luteum, especially 8IU/8 times injected in abdomen(6.33±1.63)or 8IU/2 times(6.17±1.73) in cloaca(p<0.001 v.s N/S：0.75 ± 1.5). RecFSH increased the diameter, the folding of the endometrial layer, the height of endometrial epithelium layer, the area of uterus and the area of endometrium of uterus, especially injected 8IU/4 times and 8IU/2 times. Same dose of recFSH will not change the status of ovary and uterus while injected in abdomen or cloaca. But decreasing dose of recFSH and increasing hCG in cloaca, it will reach the same result of injected in abdomen(recFSH：hCG =1：16). By the way, estrogen receptors of uterus will decrease when injected recFSH and progesterone receptors will increase. But inject recFSH and hCG at the same time, we will have different results. This finding suggested that recFSH induced follicules grow to ovulate and induced ovary mature and endometrium proliferation, especially injected the 4 IU/2 times or 2 IU/2 times dosage and supplemented hCG into cloaca is the most apparent. Furthermore, different injective site has about the same influence. Therefore, we can select low dose and low frequency (2 IU/2 times) and supplemented hCG with cloaca (eg. vagina suppository) for abate invasive therapy in the clinical apply.
