To this end, the present study was aimed at understanding its antiproliferation activity and inhibitory effect on the tumor worsen in breast cancer cells. We found that diosgenin inhibit cell invasiveness and migration in Hs578T cells through inactivation of matrix metalloproteinase 2 (MMP-2). Underlying molecular mechanism of diosgenin on PI3K/Akt pathway, the expression levels of four targeted proteins, including PI3K, Akt, GSK-3β and β-catenin were examined. In immunoblotting analysis, diosgenin-treated Hs578T cells inactivated PI3K, resulting in reduced expression of Akt, GSK-3β and β-catennin. Additionally, the dephophorylated status of Akt and GSK-3β were observed in Hs578T cells after diosgenin exposure. Furthmore, a reduced protein expression of PI3K was obtained in MCF-7 transfected with small interfering RNA (ER siRNA). Taken together, these findings provide the support that diosgenin may inhibit PI3K/Akt pathway, responding to prevent cell invasion via downregulation of MMP-2 was irrespective of ER status.
