Summary: | 碩士 === 高雄醫學大學 === 藥學研究所碩士班 === 95 === Burn is the most popular trauma in skin, and Shiunko is often used to treat the trauma for a long time in traditional medicine. The major component of Shiunko is the extract of Lithospermum erythrorhizon. Hence, chitin/PLGA film matrix containing the extract was developed as a new dosage form by wet phase inversion method in this study. The thin film composed of chitin and PLGA according various ratios by weight, the thickness of film was as low as micrometers. To evaluate the film absorptive properties, the water absorption test was performed. From the result, the film could absorb water quickly due to the increase of chitin ratio in the formulation and bring to porous characteristics. It was found that the tensile force of the film decreased as the ratio of PLGA increasing. After the morphological observation by SEM, the diameter of the pore of film was found increased as the ratio PLGA increasing. The optimal formulation was composed with the ratio of PLGA (50/50) and chitin at 1:2 in NMP or DMAc. In addition, the analysis of the shikonin and its derivative from the extract of Lithospermum erythrorhizon was performed by capillary electrophoresis (CE). In this study, CE with a phosphate buffer of pH=7.5, SDS of 50 mM, β-CD of 10 mM, 0.5% Tween 20 and 10% ACN could also successfully separate the shikonin and its derivatives.
