Summary:碩士 === 中興大學 === 土壤環境科學系所 === 95 === Medical plants have been paid more attention recently for which are more advantages, in containing more curative compositions and having less side effects, than chemical medicines. “I-Tiao-Gung” (Glycine tomentella HAYATA), has become one among the most peculiar products in Jinmen under the cooperative promotion by the Jinmen county government and local store managers. G. tomentella is a perennial legume and unable to expand its production scale because of its short growing period in a year, low germination rate and witches’ broom disease. The aims of this study were on improving its germination rate and establishing a procedure to grow healthy plantlets of G. tomentella through tissue culture. Results from the germination experiments, showed that the germination rate was increased from 25% to more than 70% and 58%, respectively, after soaking in water for 24 hours and drying at 50℃ (WD treatment) and water bath at 50℃ for 2 hours (WT treatment). The seeds can preserve for 30 days after these two treatments without loss of effectiveness. Its germination rate was increased with increasing incubation temperature after heat treatments. After a series of experiments were carried out, results showed that, the tissues of top-tip and stem-node are the highest ability in forming buds, about 90%, and the other organs, like leaf, leafstalk and stem segment could not induce any buds. The modified medium( B5S medium), with the addition of 0.5 mg L-1 BA, 0.2 mg L-1 NAA, 2% sucrose and 0.9% agar into B5 medium, can induce largest number of buds from stem-nodes of G. tomentella, reaches 3.44 buds per stem-node averagely. There was no phytoplasma found in the plantlets produced in this study, neither from the tissue culture and nor from the heat treated seeds.
