Effects of growth regulators (auxin and cytokinin) on the induction, propagation of adventitious bud, and further plant regeneration. 2. The influence of various root cutting and cutting methods on shoot propagation. 3. The propagation efficiency of tissue culture and root cutting was compared. The young leaf was the best explant resulting high rate of shoot formation, shoot number, and the lowest browning rate. The optimum condition was obtained by young leaf explants supplemented with TDZ. Except the MS medium alone treatment, the adding of any plant growth regulators (IAA, NAA, BA, kinetin, and TDZ) caused hyperhydric shoots. MS based medium contained 2 mg/l NAA leading to the highest rooting rate with 6.3 roots per shoot. Mixture with equal amount of peat and perlite was the most suitable medium for plantlet acclimatization. In consideration of survival rate and lateral shoot number, the cutting with long-thick-root was the best, followed with the long-thin-root one. Of various cutting methods, the slant insertion resulted in a higher survival rate and the straight insertion gave the highest lateral shoots. Therefore, the using of long-thin-root cuttings and slant insertion will lead to high propagation efficiency. In conclusion, a method took only 56 days from root cutting to plantlet with low cost, high efficiency, and suitable for vegetative propagation of Isatis indigotica Fortune was developed.
