Summary:碩士 === 國立成功大學 === 醫學工程研究所碩博士班 === 95 === Abstract   Collagen is a biomaterial with great biocompatibility and biodegradable which is widely used in the field of tissue engineering as the scaffold, especially for wound dressing and artificial skin. The pore size, porosity, thickness and other structure factors of scaffold would be influenced by the fabrication process. There are two objectives of the study:1. To evaluate the structure of collagen scaffold with micro computed tomography (Micro CT) approach. 2. In order to extend the application of Micro CT, establishing scanning the parameters for soft tissue with Micro CT evaluation. Micro CT sectioning have at least the follow advantages:less human effort, less time consuming, without damaging the specimen, and continuous data. The study used self-developed MatLab programs to evaluate the porosity and connectivity of the scaffold structure, along with the software provided by the Micro CT to get the 3D pore size distribution. Among the four kinds of fabrication process, each scaffold had different porosity and pore size distribution, which indicated the importance of collagen scaffold evaluation in justification the fabrication process. The results showed that 3D reconstructed from Micro CT images could be useful in the quantitative evaluation of scaffold which could further be used at the process justification.
