Summary: | 碩士 === 國防醫學院 === 生物化學研究所 === 96 === Matrix metalloproteinases (MMPs) are a group of proteinases, and they needs zinc ion for activity. MMPs have fuction in organogenesi, embryo implantation, inflammation and invasion of cancer cells. MMP-9 can promote cancer cell to invase. So it is of intrest to design three kinds of anti-cancer agents with different functional groups to chelate zinc ions, such as rhodanine, hydroxamate and rhodanine-hydroxamate. Starting from glycine methyl ester, rhodanine and rhodanine-3-acetic acid provided 25 novel compounds. After biological evaluataion, it revealed no lethal cytotoxicity under 50 mM, and compound 11e is more potent than specific MMP-9 inhibitor (ONO-4817) and broad-spectrum MMP inhibitor (GM1489).
