The restoration of subcutaneous adipose tissue remains an unresolved problem since satisfactory implant materials are still not available. Engineered tissues are being developed as alternatives. We here focus on the application of preadipocytes and injectable collagen/ hyaluronic acid gels for soft-tissue defect reconstruction. Preadipocytes isolated from the adipose tissue of male Wistar rats were cultured in collagen gels with various ratios of hyaluronic acid for in vitro and in vivo experiments. Particularly, the cell-gel constructs were evaluated with magnetic resonance imaging (MRI) after subcutaneous injection. We found the addition of hyaluronic acid in collagen gels increased proliferation of the preadipocytes, and limited contraction of the gels. Mature adipocytes were observed in the cell-gel constructs after subcutaneous injection by histological analysis. These results support the promise of preadipocytes combined with collagen/ hyaluronic acid gels as a material for use in producing soft tissues in vivo.
