Organogenesis requires the increases of cell size and cell number as well as the support of vascular system. VEGF, a selective mitogen for vascular endothelial cells, is important for the development of vascular system. But it is not clear if PTEN controls tissue mass by suppressing angiogenesis provoked by VEGF and their correlations on the postnatal tissue development in fish. We used a primitive vertebrate, eel, to study this question. Two forms of PTEN (PTEN Long- or Short- form Like) cDNA have been cloned in Japanese eel. The eel’s cells were treated with IGF-1 and Troglitazone to investigate the expression control of PTEN in vitro. The results showed that IGF-1 suppressed while Troglitazone stimulated PTEN expression. Rete mirabile,ovary, and Liver from normal eels or eels injected with hypophysial factors were used to investigate the expression of VEGF A, PTEN, IGF-1, and Flk-1 by RT-PCR in vivo. There was a negative correlation between PTEN expression and ovarian tissue mass and a positive correlation expression with VEGF and a higher correlation between the tissue mass and the ratio of VEGF to PTEN. There was a negative correlation between PTEN expression and RMI and a positive correlation expression with VEGF. VEGF expression was suppressed in a later period on the Rete mirabile, but it was still a higher correlation between the tissue mass and the ratio of VEGF to PTEN, so the development of tissue to seem to display the main factor with PTEN expression. We showed that the involvement of VEGF and PTEN in the postnatal tissue development, the ratio of VEGF to PTEN may represent the growth status of the specific tissue.
