Summary:碩士 === 長庚大學 === 化工與材料工程研究所 === 96 === In this research﹐we use the supercritical carbon dioxide dynamic extraction of resveratrol of Polygonum cuspidatum﹐and the extractions were analyzed by High-Performance Liquid Chromatography﹒Our purpose is to separate the highly value-added resveratrol from the Polygonum cuspidatum with a absolutely pure extraction facility﹒In order to find the best operation condition﹐various factors such as extraction temperature﹐extraction pressure﹐different modifiers﹐rate of flow﹐extraction time﹐and the holding time were discussed﹐Furthermore﹐the comparisons between the productions of traditional extraction and supercritical fluid extraction have been made﹒ The results show the intention substance is high extraction efficiency in 70 ℃﹐5000 psi﹐and 6 hours﹒Besides we test methanol﹐ethanol﹐and water as a modifier﹒However the ethanol shows the highly percentage of extraction and its safe for people﹒ A reversed HPLC method was applied﹐and the best proportion for mobile phase is water:acetonitrile=74:26 with 0.9 ml/min flow rate﹒The maximum absorbability wavelength is at 303 nm wavelength﹒
