The specific primers were designated to amplify the target DNA fragments from pig genomic DNA by PCR. PCR products were then purified to process dideoxy single-base extension reaction. The extension products was detected on the capillary electrophoresis and analyzed by software for genotyping by minisequencing. PCR-RFLP or direct sequencing was also performaned for the control. Genotypic and allelic frequencies of each gene in Landrace, Duroc, Yorkshire, LD and black pigs were analyzed. It was found there were existences of alleles are favored in L-FABP, CCKAR, ADSS2, MYF5 and CAPZ genes. In the contrast, HUMMLC2B and TXNIP genes could be improved in these pig breeds. Further study was carried out to investigate the correlation between genotypes and performances in pigs by SAS statistical analysis. The average daily gain of TT type of HUMMLC2B gene was significant higher than TC and CC types in Duroc. The backfat of TT type of ADSS2 gene was thinner than the TC type. The CC type of TXNIP gene was also thinner than the TT type in the black pigs. The carcass length of CC type of HUMMLC2B gene was the longest in black pigs. The TT type of TXNIP gene in LD and NN type of MYF5 gene in black pigs were longer than other types, while TT type of ADSS2 gene was significant shorter than TC and CC types in LD and black pigs. The rib numbers of CC type of HUMMLC2B gene was the most in LD. All SNP genotypes of those genes could be detected by multiplex minisequencing in a same capillary.
