In this study, the safety of fresh Polygonum multiflorum Thunb was concerned, therefore that was assessed the safety and antimutagenic activity. The first, the root, stem and leaf carried on the extraction by the water extraction of supersonic wave to process under experiments. The results showed that the water extracts of root, stem and leaf of Polygonum multiflorum Thunb. at 0.1~1.0 mg/plate did not only show any cytotoxicity and mutagenicity on Salmonella typhimurium TA 100 and TA 102 by Ames test, but also not showed the cytotoxicity for CHO-K1. Therefore, in sisters chromosomic exchange frequency assay, there is not significance difference (p<0.05) between control and sample. The water extracts of root, stem and leaf of Polygonum multiflorum Thunb. represented the antimutagenicity and bio-antimutagenicity influences. The root water extracts was the greatest on suppressed mutagenicity activity for direct mutagen (N-methyl-N’-nitrosoguanidine) or indirect mutagen ﹝benzo(a)pyrene, which respectively reached 31.1% and 74.3%. There is a demonstration that the water extracts of root, stem and leaf from Polygonum multiflorum Thunb. were expression better antimutagenicity activity, and for secure raw material. The active ingredients of fresh Polygonum multiflorum Thunb on promotion antioxidative properties and intracellular antioxidant enzymes activity have been confirmed in this study. The results showed that the total phenolic of the water extracts of root, stem and leaf of Polygonum multiflorum Thunb respectively were 179.0, 70.3 and 118.1 mg/g, but flavonoids respectively were 7.3, 8.0 and 37.5 mg/g. In respect to the antioxidative capacity assay, that display the best capacity of scavenging DPPH and NO&;#8226; free radical (90% and 35 %) with water extracts from root of Polygonum multiflorum Thunb. The reducing power and TEAC assay both also took root water extracts as best, but the scavenging superoxide anion of extracts from stem and leaf more than root extraction. Additionally, the water extracts from root of Polygonum multiflorum Thunb. could promote intracellular GSH levels cause its obvious to increase the activities of glutathione peroxidase, glutathione reductase and glutathione-S transferase intracellular for Clone 9 cells. The functional constituents of fresh Polygonum multiflorum Thunb from water extraction were analyzed, the extracts of the root contained lot of emodin (6.6 mg/g), but the major functional component of leaf water extracts is qucertin (13.5 mg/g). The fresh Polygonum multiflorum Thunb water extracts unfolded the optimization antioxidative activity under lower concentration, that appeared emodin and quercetin were mainly active constituents. Epidemiology indicated that antioxidants qualified to decrease affection cancer hazard. Some studies also showed native antioxidants could expression cytotoxicity for cancer cells. Further, so we confirmed the root water extracts of fresh Polygonum multiflorum Thunb induced apoptosis of mechanism for Hep G2 cells. Results showed the extracts could effective to decrease Hep G2 cells survival and that appeared time-dependent manner. Another, the extracts can inducible Hep G2 cells accumulated on subG1 phase (8.33 %) and involved intracellular ROS production (fluorescence intensity up to 154.4, % of control). In annexin V-FITC/PI double stained assay, the cell apoptotic ratio would be respectively increased to 78.2 or 77.4 % after cells were pretreated with 24 or 48 h, however, the activity of caspase-3, -8 and -9 also were stimulated. From above results, we could know the water extracts of fresh Polygonum multiflorum Thunb can suppress Hep G2 cells proliferation and induce apoptosis. The hepatoprotections of the water extracts from root of Polygonum multiflorum Thunb.(RPW) against CCl4-induce liver damage on rats has been studied. Therefore, we used carbon tetrachloride (CCl4) as liver damage inducer and explained protection effects of RPW direct at CCl4-induced liver damage in a model of rats. Animals were fed the extracts of RPW for eight weeks. Our results showed that GOT and GPT levels had significant differences (p<0.05) between negative control and blank, but positive control (silymarin, 200 mg/kg b.w.) and sample (feed 200 mg/kg b.w. and 400 mg/kg b.w. RPW) are both appearing significant to decrease GOT (74, 85, 88 %, respectively) and GPT (38, 66, 70 %, respectively) levels of serum. Additionally, the RPW could promote GSH levels cause its obvious to increase the activities of glutathione peroxidase, glutathione reductase and glutathione-S transferase in liver tissue when compared with CCl4-treated group (p<0.05) to appear antioxidative capability. The results of this study show that a RPW diet can be proposed to protect the liver against CCl4-induced oxidative damage in rats. Farther, we investigated some cytokines production about inflammation or liver-damage includes IL-18 and TNF-α. Results showed a RPW diet direct significantly (p<0.05) lowered the IL-18 and TNF-α level of inflammation maker in serum. Therefore, RPW could expression effects of inhibited CCl4-induced liver damage or suppressed inflammation, the hepatoprotective effect might be correlated with its antioxidant activity. To integrate above results obtained a conclusion that fresh Polygonum multiflorum Thunb has antioxidative and anticancer effects that can used to be the herbal medicine, which has potential of the health products development.
