The study I contained 2 experiments for the efficiency of oocytes collection method and the subsequent development to blastocyst after in vitro maturation (IVM) and in vitro fertilization (IVF). It was resulted that the slicing method could obtain significantly more cumulus-oocyte complex (COCs) in good quality compared to aspiration method (55.7% vs. 33.0%, P < 0.05), although there was no significant difference in the oocytes developed to MII stage after IVM in two methods (78.3 vs. 77.7%, P > 0.05) (Study-Ⅰ, experiment-1). When the COCs collected with slicing method and by IVM-IVF and subsequently cultured in the synthetic oviduct fluid solution (SOF) medium or co-culture medium with cumulus cell dividedly, both the cleavage rates and blastocyst rates were similar(28.2 and 37.1 vs. 4.3 and 0.0%, P > 0.05). However, only the COCs cultured in SOF medium had the chance to break out of the block stage and develop to blastcyst stage(Study-Ⅰ, experiment-2). The study Ⅱ is aimed to valuate the effects of the supplements of cysteamine and/or epidermal growth factor (EGF) into the usual IVM medium (control) on the development rate of in vitro culture (IVC) after IVM-IVF, and to compare the development rate of blastocyst obtained by IVF using fresh and frozen semen. Results showed that the blastocyst rate of the IVM medium in the supplement of cysteamine combined with EGF was significant higher than the other two groups (36.1 vs. 15.7 and 10.0%, respectively, P < 0.05)(Study-Ⅱ, experiment-1). Besides, the oocytes cultured in the higher effective IVM medium formerly selected had almost same cleavage rate (87.6 vs. 88.0%) and blastocyst rate (24.6 vs. 26.0%) after IVF comparing with fresh and frozen semen(P > 0.05)(Study-Ⅱ, experiment-2). Experiment in the study Ⅲ is to examine the difference of age in blastcyst formation rate of the oocytes after IVM-IVF and IVC in the post-pubertal young and multiparous goats. It was found that the oocytes obtained from young goats had higher capability in the development of blastocyst than the goats with multiparity (27.5 vs. 14.1%, P < 0.05). In couclusion, it is suggested that the COCs collected using slicing method from young post-pubertal goats, and the supplement of cysteamine combined with EGF in the medium could obtain more high quality matured oocytes. Furthermore, it has advantage of higher blastocyst production rate of the matured oocytes in vitro fertilized with ether fresh or frozen semen, and subsequently cultured in the SOF medium.
