In this study, we sampled pigeons from 195 farms over the 2007 years. The young showed low hatching rate and it had signs of lethargy, weight loss, respiratory distress, diarrhoea and poor racing performance and a variety of other signs. Histological features of PiCV infection were the presence of basophilic intracytoplasmic inclusion bodies (19/195; 9.7%). Based on pathologic studies,the bursa of Fabricius was the main target organ. Using PCR for the detection of PiCV in infected pigeon, the expected 206-bp products were obtained with a set of degenerated primer targeting Capsid gene for PiCV, and 15 cases showed positive reaction (15/195; 7.6%). PiCV infections were mainly found in the pigeons under 4 month of age in this study. Using the DNASTAR software to analyze protein amino acid sequences, our isolates could be further divided into 3 clusters ( A to C ). The most of the strains ( 10/15、4/15) were belonged to cluster A、B. The cluster were similar to Belgium (DQ915957 and DQ915958 ). Cluster C (1/15) was identical to USA (DQ915961 and DQ915962 )、France strains (DQ915960) and Czect strains (AY461810 and AY887540 ). The clusters A and B had no identical sequences deposited in NCBI. It might be as a local strain with unique genotype in pigeon in Taiwan. Among the three genotypes, type A is the most predomineat and often associated with secondary infections. Using in situ hybridization, bursa、thymus、spleen、gut-associated lymphoid tissues、bronchus-associated lymphoid tissues、nasal cavity、crop and eye showed intense PiCV nucleic acid labelling of cells.
