Summary:碩士 === 慈濟大學 === 醫學生物技術研究所 === 96 === Abstract This study developed a method capable of direct quantitation of 6-acetylmorphine and oxycodone in urine sample by preparing capillary immunoaffinity columns (CIACs) and applying the CIACs to a liquid chromatography electrospray ionization - mass spectrometry system (LC/MS). CIACs were prepared by packing anti-morphine mouse monoclonal antibody immobilized silica into capillary columns (I.D. 100 μm, O.D. 365 μm and length 150 mm). Two CIACs were used for extracting analytes from urine sample and detecting the analytes by a LC/MS system alternatively. Aqueous ammonium acetate buffers were used as the mobile phase. Urine samples spiked with different concentrations of 6-acetylmorphine (6-AM) or oxycodone were added with fixed concentrations of 6-acetylmorphine-D6 (6AM-D6) or oxycodone-D6 as internal standard (IS), filtered through a membrane (0.22 μm), and injected into the LC/MS system without any further pre-treatment. The limits of quantitation and the limits of detection were 15.5 and 4.6 ng/mL for 6-AM and 39.0 and 11.7 ng/mL for oxycodone. And precisions ranged from 1.0% to 2.9% for 6-AM and 3.0% to 7.6% for oxycodone. These results exhibited that accuracy and precision (n=3) of the method are fairly good.
