The concentration of procaterol present in human plasma is sometimes even lower than the detection limit of analytical instruments commonly used. So far, only GC/MS and radioimmunoassay can meet detection requirements. In this study, the pretreatment of samples by extraction in combination with the high performance liquid chromatography coupled with electrospray ionization tandem mass spectrometry was used to quantitate the concentration of procaterol in heparin-containing human plasma. Good linearity of 0.005-1 ng/mL with coefficients of determination which was not less than 0.998 was obtained. The signal to noise ratio (S/N) for the lower limit of quantitation (LLOQ) 0.005 ng/mL, was approximately 11, which is higher than that reported by Tu et al. (2005). The peak area ratio of procaterol at LLOQ and blank plasma at the retention time corresponding to procaterol was approximately 19. These results clearly indicated this quantitative method has excellent stability and reliability. The validation analyses of within-run and between-run demonstrated that the accuracy (relative error, %RE) for LLOQ was not more than 20.0% while those for other procaterol concentration examined was not more than 12.0%. The precision (coefficient of variation, %CV) for LLOQ wasdetermined to be between -16.8% and 8.8% while those for other procaterol concentration examined was between -3.0% and 8.7%.
