Callus regenerated into adventitious roots gradually during cultural period. Total flavonoid of methanolic extract in adventitious roots was 12.99±1.55 eq. iridin % DW. Five kinds of flavonoids (tectoridin , iridin , tectorigenin , isorhamnetin and irigenin ) in methanolic extract were analyzed by HPLC. Mature adventitious roots (> 3cm) were cultured in MS liquid medium supplemented with L-phenylalanine or yeast extract produced high isoflavonoid(3.8~4.1 % DW). For scale up, adventitious roots were cultured in 1L bubble column bioreactor in 500 ml MS liquid medium for 4 weeks resulted 3.6-fold increase in biomass. The methanolic extracts in adventitious roots cultured in bubble column bioreactor exhibited 1.3-fold in total flavonoid and 1.2-fold in total isoflavonoid, compared to 3-year-old medicinal rhizome.
