Summary:碩士 === 元智大學 === 生物科技與工程研究所 === 96 === According to reference, fungi has better ability of producing cellulases and hydrolysing cellulose to reducing sugar, then converting those sugar to ethanol.Therefor,this study focus on fungi hydrolyze cellulose to reducing sugar.Based on Aspergillus niger can hydrolyze cellulose,trying to find optimal condiction for Aspergillus niger and get higher efficience to hydrolyze cellulose to sugar.The factor that affect the enzymatic hydrolysis of cellulose include substrates, cellulase activity, and reaction conditions (temperature, inoculum volumn, agitation speed, and other parameters). In order to improve the reducing sugar yield and hydrolysis rate, this research is focus on optimal hydrolysing condition and medium component, which can increasing the cellulase activity and then improving the yield and rate of cellulose hydrolysis. This research demonstrated that the best condition of temperature, inoculum concentration and the rotational speed is 30℃, 3.6×1010 spore and 100 rpm. The better nitrogen source is soytone. At this condition, the reducing sugar weight and yield are 36.4% and 364ppm.
