Consequently, anti-cancer drug market and the demand have also gradually been increasing accordingly. One research in this area is to find more environmental friendly methods or green manufacturing process for the production of anti-cancer drug. This experiment uses Taiwan's native yew Taxus sumatrana as the for the extraction of taxol and 10-deacetyl baccatin III (10-DAB) by pressurized water extraction (PWE). High performance liquid chromatography (HPLC) equipped with a carbon-18 column was used for the quantitative determination of the extracts. The results of PWE were compared to conventional organic solvent extraction. Experimental conditions including leaves crushing, pH of extraction, extraction pressure, mixing of the sample with sand, and static extraction time were studied. Crushing of the leaves and static extraction time were found to be the more influential parameters. For taxol, our results showed that 31 mg taxol can be obtained from one kg of dry leaves, while 1698 mg 10 - deacetylation baccatin III can be obtained from one kg of dry leaves.
