Orf virus can infect goats and sheep, causing proliferative lesions around the lips and nostrils. Orf is endemic in Taiwan, with low mortality and economic loss is not high. In this thesis, first, the complete sequence of the B2L gene of orf virus from an outbreak in Nantou was determined and the phylogenetic relationship of the B2L gene with other strains was compared. Second, the 3’- terminal heterogeneity of the A32L gene was found from analyzing sequence data of the A32L gene of several orf outbreaks. Among the three viral genes (B2L, E3L and A32L) studied, the B2L and E3L genes showed single nucleotide changes; however, the A32L gene showed heterogeneity in the 3’-terminal region, including nucleotides variation and deletion. Third, a PCR method of the A32L gene was developed for rapid diagnosis of the orf virus infection; our method can distinguish different orf strains in a single-step PCR reaction.
