This experiment uses Chinese hamster ovary cells (CHO cells) to analyze the transfection efficiency of eukaryotic expression of porcine IL-18 plasmid, and uses the 4 weeks old specific-pathogen free (SPF) pigs to test it and take it as the adjuvant benefit of Japanese encephalitis virus (JEV) DNA vaccine. The result demonstrates that the growing amounts of the splenocytes IL-12 and IFN-γ from the animals injected with IL-18 adjuvant are higher than the pigs that are only injected with JEV DNA vaccine. Moreover, the gene expression of IFN-α and TLR3, 7 and 8 increase yield in the animals injected with IL-18 adjuvant. Therefore, eukaryotic expression of porcine IL-18 plasmid owns the ability to adjust the immune effect of the JEV DNA vaccine that is injected at the same time, which can effectively improve the function of the vaccine.
