Chlamydophila abortus is able to colonize the placenta of several species of animals and which may induce abortion. C. abortus is the most common cause of infectious abortion in sheep and goats in the Taiwan, resulting in major economic losses. Chlamydiosis is an important zoonosis and several cases in ruminants and birds have been reported in Taiwan. Diagnostic kits imported from other countries are routinely used herein but their high costs are still a major problem. Therefore, it is necessary to develop an effective and economic diagnostic kit for local utility. The BALB/c mice were immunized with purified protein combine adjuvant for four times with each interval of two weeks. Then fuse BALB/c mice spleen cells and NS 1 cells and to produce monoclonal Abs. In western blotting experiments shows the mAbs have the specificity to VD1 protein. The isotyping of all the mAbs (SD1~SD8) are IgG1 heavy chain and κ light chain. The SD1 mAbs can recognize the specific antigen in C. abortus infected placenta tissue section by the Immunohistochemistry test. To understand the sensitivity of competition ELSA in goat serum and goat Swap by SD1 mAbs. Use competition ELSA to test the goat serum and goat Swap samples. In the future, we will identify the recognition epitope and detection sensitivity of all the mAbs.
