The clinical symptoms, including high fever, vomiting, and lethargy, are not disease-specific and the bacteria that associate with adult bacterial meningitis are geographically different.Therefore, it is important to develop a rapid and accurate methodology for clinical diagnosis. Current methods for detecting bacteria and bacterial components in cerebrospinal fluid, such as polymorphonuclear cells, Gram stain, latex agglutination(LA), acridine orange stain, and quellung capsular reaction, are not sensitive and specific enough. In addition, they are easily interfered with cross-reaction. The present study aimed to develop a highly bacteria species-specific and rapid method for the diagnosis of acute bacterial meningitis. First, we modified the DNA extraction procedure from QIAGEN commercial kit and established a mutiplex-PCR condition to target the DNA of different bacterial strains. Then, we evaluated the detection sensitivity of optimized mutiplex-PCR for these strains.
