Description
Summary:碩士 === 長庚大學 === 生物醫學研究所 === 98 === Diabetes mellitus(DM) is the most prevalent endocrine disease in developed countries. Increased vascular permeability and pathological neovascularization are the prominent features in the diabetic retinopathy. It is generally believed that the development of these pathological processes is closely correlated with the chronic hyperglycaemia. In our previous study , we demonstrated that resveratrol possesses hypoglycemic effects in streptozocin-induced DM ( STZ-DM ) rats. Hower , the possible effects of resveratrol on the anti-retinopathy in STZ-DM rats have yet to be examined. The purpose of this study is to invegstigate the effects of resveratrol on retinopathy in STZ-DM rats. Vascular permeability and neovascularization in retina of the STZ-DM rats were determined. The possible effect of resveratrol on the signal pathways responsible for the retinal neovascularization was also examined. We showed that resveratrol attenuates DM-induced retinopathy in STZ-DM rats. In resveratrol-treated DM rats , the vascular permeability and neovascularization were significantly attenuated. This reduced retinal vasculopathy was correlated with the decreases in STAT3 activity and VEGF production by rsveratrol in the retinal tissue. However , resveratrol seems to modulae the inflammatory state of the diabetic retina via NF-κB pathway, but this experiment is not confirmed yet.
