Summary: | 碩士 === 長庚大學 === 生物醫學研究所 === 98 === Celastrol is one of active compounds in Thunder God Vine (Tripterygium wilfordii Hook F.) root, and often used as a remedy of inflammatory. Also, celastrol may affect the tumor cells through apoptosis, reduced cell cycles and growth. Interleukin (IL)-6 is produced at high levels by RCC and plays an important role. The effect of celastrol on renal cell carcinoma (RCC, 786O and A498) is still unclear. This study is to investigate the effect of celastrol on IL-6 secretion from RCC. A498 and 786-O cells were cultured in RPMI-1640 with different concentrations of celastrol at different time periods. The media IL-6 concentrations were measured by enzyme-linked immunosorbent assay (ELISA). The expression of IL-6 mRNA was determined by reverse-transcriptase polymerase-chain reaction (RT-PCR). The phosphorylated ERK, JNK, and p38 proteins were determined by western blot. The cell cycle of RCC was measured by flow cytometry. The results showed that: (1) reduced media IL-6 secreted from RCC treated with celastrol was observed; (2) PD98059 (MEK inhibitor) reversed the inhibitory effect of celastrol; (3) attenuated IL-6 mRNA expression was found in RCC treated with celastrol; (4) inhibited IB protein and induced phosphorylation of ERK were found in RCC treated with celastrol; (5) reduced proliferation of high concentration of celastrol was observed in RCC cells by MTT assay; (6) celastrol also causes cell cycle arrest at G0/G1 phase. In conclusion, the inhibitory effect of celastrol on IL-6 secretion is, at least in part, through the elevated ERK phosphorylation, reduced IB, attenuated expression of IL-6 mRNA, and reduced cell proliferation. Also, celastrol causes cell cycle arrest on RCC cells.
