Summary:碩士 === 長庚大學 === 中醫學系天然藥物 === 98 === Daphne genkwa Sieb. et Zucc. is one kind of Daphne plants (Thymelaeaceae). It was distributed in Europe, Taiwan and China. It has been used as a curative from number of ailments, such as anti-tussive, anti-tumor and anti-inflammatory. In the preliminaryest, the chloroform extract of D. genkwa showed 61.5 % inhibition against A549 at 30 μg/mL. Therefore, we were interested in the bioactive principles of D. genkwa. The dry flower buds (5 Kg) of D. genkwa were extracted by methanol. The filtrate was concentrated under reducing pressure to give dark brown extract. The crude extract was suspended in water and partitionated with chloroform, n-BuOH, separately. Each layer was analysed by column chromatography, thin layer chromatography (TLC) and high performance liquid chromatography (HPLC) to isolate pure components. Forty-five compounds were isolated from the flower buds of D. genkwa. Their structures were determined by spectral analysis and compared with literature data. The major compounds are flavonoids and lignans. Their bioactivities are under proceeding. Four compounds were isolated for the first time from natural products.
