After weanling, Sprague-Dawley rats were fed riboflavin deficient diet ad lib (D), riboflavin adequate diet ad lib (A), or riboflavin adequate diet pair-fed (P). Animals were s.c. injected with 100 mg/kg D-galactose (DG) daily till 16 wks of age for inducing ageing. The riboflavin adequate-natural aging control animals (N) were given phosphate buffer saline (PBS) s.c. during the study. Three hours before being sacrificed, half of the animals received i.p. injection of diquat to induce their oxidative stress. Riboflavin deficiency of rats was confirmed by a significant increase in EGRAC after 2 weeks. DG injection increased GR activity. The oxidative stress indicators showed no significant difference between groups. Hepatic total glutathione were significantly higher in D group than in P group. Hepatic Mn-SOD and CuZn-SOD activies were significantly increased only in D group. GR, CAT and GPx activies were not significantly changed. There were no significant difference in Mn-SOD and GR protein expression. When treated with a dose of diquat without causing liver damage, an increasing trend in plasma and hepatic TBARS were observed. Under the oxidative stress, hepatic GR was significantly increased in P group but decreased in A group respectively. Hepatic total glutathione, GSSG and GSH were significant increased only in D group. CuZn-SOD activity was significant higher in D group than in P group. Mn-SOD, CAT and GPx activies were not significantly changed. Mn-SOD protein expression was significant higher in D group than in A group. GR protein expression was not significantly different. Long term DG injection since young age neither enhance oxidative stress nor resulted in increased aging of rats. On the other hand, riboflavin deficiency may result in the compensatory increase in the antioxidative enzyme activities and protein expression under high oxidative stress.
