In the modify process, the quality of growth of Self -assembled monolayer (SAM) dominates the response of detection. Since the thickness of monolayer are only a few nanometers, we want to ensure that modify process is fully function. This study utilizes Surface Plasmon Resonance (SPR), Fourier Transform Infrared (FTIR) and Contact Angle (CA) to observe the forming process of the SAM on the Au film. Through high-resolution SPR angle scanning, the variation of resonant angle due to modify process is obtained. FTIR and CA are used to measure the difference of molecule bonding oscillation and hydrophobic of surface modified before and after, respectively. In this study, we etch Pitch 1.3μm diffraction grating on a CD-like polycarbonate substrate and coat Au film which thickness is about 50nm. The Au film surface of Grating find application in biosensor then interact with bio-molecule on specific functional group surface by select modify method. Besides, we distinguish the alkane chain length by selecting the material: 16MHDA, 8MOA and 3MPA. Utilize carboxylic group and Mercaptan for terminal and ligand, respectively. We find the most stable modify process by measuring the COOH-terminated alkanethiols on Au film and activated yield. In result of SPR measurement, we concluded from the direction and quantities of angle shift are consistent with the simulation results of Gsolver and experimental value. In the same condition of Terminated activated step, 16MHDA shows the best performance. In result of FTIR measurement, 16MHDA, 8MOA and 3MPA represent the same characteristic peaks. In result of CA measurement of surface, the hydrophobic order would be 8MOA, 16MHDA and 3MPA, respectively. In this study, we obtain a more stably modifying process and activating process with COOH-terminated alkanethiols on Au film.
