The discovery of skin aging mechanism reveals that skin aging are related to free radical production and chronic inflammation .This study was to investigate the antioxidative activities of ten Chinese herbal medicine extracts and their inhibitory effects on nitric oxide(NO) production in lipopolysaccharide (LPS)-activated macrophages. The ten Chinese medicines are Lilium brownii.(百合), Forsythia suspensa(連翹), Coptis chinensis Franch.(黃蓮), Gardenia jasminoides Eills.(梔子), Fructus kochiae Scopariae.(地膚子), Lithospermum erythrorhizon Sieb.(紫草), Santalum album Linn.(檀香), Artemisia argyi.(艾草), Crataegus pinnatifida Bunge.(山楂) and Coix lacryma-jobi.(薏仁). Among them, Forsythia suspens and Sandalwood had good performance on scavenging DPPH radical (IC0.20:15.8±7.1 and 21.1±3.2 μg/mL, respectively ). By comparison, Lilium brownii and Coix lacryma-jobi had poor performance on scavenging DPPH radical (IC0.20> 700 μg/mL ). Sandalwood and Artemisia argyi Levl significantly suppressed nitric oxide(NO)production concentration-dependent in lipopolysaccharide(LPS)-activated macrophages with IC50 11.6±2.9 and 16.4±6.7 μg/mL, respectively.It was found that the partial purified fraction 6, 4 and 5 of Sandalwood also showed suppressive effects on NO production in LPS-activated macrophages with IC50 13.6±1.3, 54.9±5.4 and 30.7±11.0 μg/mL respectively. In the cell viability assay by MTT methods showed that the partial purified fraction 1 may have cyto-toxicity effect. Western blot analysis indicated that the partial purified fraction 4 and 5 of Sandalwood extract showed significantly inhibitory effects on iNOS expression but not COX-2 expression. The future work is to identify the antioxidative ingredients, which can inhibit NO production in LPS-activated macrophages, in these Chinese herbal medicines.
