Wolbachia can infect many animals including filarial worms, crustaceans, spiders and insects, and influence host reproductive behavior, such as cytoplasmic incompatibility, parthenogenesis and male feminization. This study investigated the presence of Wolbachia in Idioscopus niveosparsus, Idioscopus clypealis, Kolla paulula and Empoascanara sonani. And, using cell culture technology culture the cells of leafhoppers for Wolbachia biology study. The results showed that I. clypealis, I. niveosparsus, K. paulula and E. sonani are all infected with Wolbachia. Phylogenetic relationships analysis results showed that I. clypealis, K. paulula and E. sonani have Wolbachia strains belonging to A- and B- supergroups, but I. niveosparsus only contains a Wolbachia strain belonging to A- supergroup. Male and female K. paulula , but not I. clypealis, I. niveosparsus, E. sonani cells could be cultured in DMEM medium supplemented with 20% fetal bovine serum. The doubling time of K. paulula cells was 8~10 day. Both male and female K. paulula cells also contain Wolbachia.
