In previous studies, it had been verified that curcumin had lots of pharmacological activities, including antioxidant, antimicrobials, and anti-inflammation. Recent researches pointed out curcumin have anti-inflammatory effects because it would inhibit the expression and activities of NF-κB. Besides, curcumin had frequently been used as a drug for chemoprevention. Previous studies also found that curcumin could modulate proliferation and apoptosis of various cancer cells. In this study we used a human monocyte THP-1 cell line as a model to investigate whether curcumin can induce apoptosis via p53-independent pathway. The results exhibited apoptosis-related protein expression would be increased after curcumin treatments. On the other hand, curcumin treatments also affected Akt and JNK signaling transductions; even more affected a group of transcriptional factors, FoxO family, in compartmental distribution between nucleus and cytosol. When using SP600125 (JNK specific inhibitor) block JNK pathway, caspase-3 activity was decreased. These results suggested that JNK play an important role in curcumin-induced THP-1 apoptosis.
