In this study, hypoglycemic activity was detected in hot water extracts of Polygonatum ordoratum(Mill.)Druce. ICR-system mice were induced diabetes by using streptozotocin ( STZ ). Animals showing blood glucose levels 250~300 mg/dL 48 h after STZ treatment were selected for study. In STZ-diabetic model, mice were treated with 50, 200, 300 mg/kg hot water extracts of P. ordoratum, respectively for 14 days. And then the fasting blood glucose levels were detected and analyzed compared with the STZ-diabetic mice group. The Glybenclamide was used as a positive control drug. According to the preliminary results, in STZ-diabetic mice model, glucose concentrations were increased in serum of diabetic animals in comparison to normal mice. The blood sugar was improved( p<0.05 ) after treatment with 50, 100, 300 mg/kg hot water extracts of P. ordoratum and the value was 429.8 ± 36.4, 307 ± 141, 286 ± 68 mg/dL, respectively. In addition, 200 mg/kg hot water extracts of P. ordoratum demonstrated the best Triglyceride( 73.2 ± 26.8 U/L ) decreased effect ( p<0.05 ). Based on the results of this study, P. ordoratum(Mill.)Druce is indeed to regulate blood sugar. The feed data is record in this study, cause one of the symptom of diabetes mellitus is eat too much. Based on the results, In the second day, In STZ-diabetic model, mice were treated with 50, 100, 300 mg/kg hot water extracts of P. ordoratum(Mill.)Druce, feed value was 41, 42, 42 g, are lower than STZ-diabetic mice 63 g. In the eight day, feed value was 52, 62, 55 g, are more lower than STZ-diabetic mice 94 g. Based on the results, hot water extracts of P. ordoratum(Mill.)Druce can control the feed amount. About In vivo anti-oxidation, experiments by determination of liver lipid peroxidation, after the results showed that: oral vote with the hot water extract of P. ordoratum(Mill.)Druce 50,100 and 300 mg / kg in mice whose TBARS concentration ( 40.4 ± 2.0, 38.4 ± 0.4, 40.7 ± 0.8 nmole / μg protein ) than those in STZ-induced group ( 49.4 ± 2.0 nmole / μg protein ) in diabetic mice with low ( p <0.05 ), another protein in the liver test results showed that: oral vote and the hot water extract of P. ordoratum(Mill.)Druce 50,100 and 300 mg / kg in mice the protein content (36.32 ± 0.31, 40.74 ± 0.80, 37.51 ± 0.48 mg protein / ml) than those in STZ-induced group ( 30.96 ± 0.88 mg protein / ml ) of the diabetic mice of high ( p <0.05 ), while in Glutathione peroxidase ( GPx ) activity assay results showed that: oral vote with the hot water extract of P. ordoratum(Mill.)Druce 50, 100 and 300 mg / kg in mice and its activity as a ( 3.49 ± 0.04, 2.79 ± 0.04, 3.25 ± 0.05 unit / mg protein ) than those in STZ-induced group ( 4.11 ± 0.14 unit / mg protein ) in diabetic mice with low ( p <0.05 ). From the above results confirm that P. ordoratum(Mill.)Druce have antioxidant effects. Tang-Yao Hong Den-En Shieh 洪堂耀 謝登恩 2010 學位論文 ; thesis 76 zh-TW
